چکیده
Lutheran glycoprotein (Lu gp) has five predicted immuno4, and Lu13 and Au to domain 5. The organization of the LU gene was determined. The region encoding the open globulin superfamily (IgSF) domains. K562 cells were transfected with Lu cDNA and tested by flow cytometry with reading frame is arranged in 15 exons extending over ̧11 kb on chromosome 19q13.2. The Lu/Lu and Au/Au blood monoclonal antibodies and Lu blood group antisera. The results confirmed the identity of Lu cDNA. Deletion mutants group polymorphisms were studied using genomic DNA from typed blood donors. The Lu mutation is a base change lacking the regions encoding one or more IgSF domains were made by inverse polymerase chain reaction (PCR), exin exon 3 (G to A) encoding an Arg (Lu) to His (Lu) change on the CFG face of domain 1. The Au/Au polymorpressed in K562 cells, and tested with the same antibodies. The Lu and Lu5 antigens and the epitope recognized by phism is an A to G substitution in exon 12 encoding a Thr (Au) to Ala (Au) change on the G strand of domonoclonal antibody BRIC 224 were mapped to the first, Nterminal, IgSF domain. Lu4 and Lu8 were mapped to domain main 5. q 1997 by The American Society of Hematology. 2; Lu20 to domain 3; Lu7 and BRIC 221 epitope to domain

Polymorphic analysis of the Lutheran blood group system in Chinese.
The Lutheran blood group is a complex system consisting of 19 identified antigens. It belongs to the immunoglobulin family of receptors and adhesion molecules. Four pairs of antigens show allelic relationships while other antigens are of very high incidence. In this study, we performed genetic polymorphism analyses by molecular techniques of the Lutheran blood group system in Chinese subjects. ...
متن کاملAn Antibody to the Lutheran Glycoprotein (Lu) Recognizing the LU4 Blood Type Variant Inhibits Cell Adhesion to Laminin α5
BACKGROUND The Lutheran blood group glycoprotein (Lu), an Ig superfamily (IgSF) transmembrane receptor, is also known as basal cell adhesion molecule (B-CAM). Lu/B-CAM is a specific receptor for laminin α5, a major component of basement membranes in various tissues. Previous reports have shown that Lu/B-CAM binding to laminin α5 contributes to sickle cell vaso-occlusion. However, as there are n...
متن کاملAn Anti-Human Lutheran Glycoprotein Phage Antibody Inhibits Cell Migration on Laminin-511: Epitope Mapping of the Antibody
The Lutheran glycoprotein (Lu), also known as basal cell adhesion molecule (B-CAM), is an Ig superfamily (IgSF) transmembrane receptor for laminin α5. Although Lu is not present in normal hepatocytes, its expression is significantly increased in hepatocellular carcinoma (HCC). In this study, we isolated thirteen phage antibodies to Lu from a phage library of peripheral blood from HCC patients, ...
متن کاملA unique gene encodes spliceoforms of the B-cell adhesion molecule cell surface glycoprotein of epithelial cancer and of the Lutheran blood group glycoprotein.
Two new members of the Ig superfamily, the Lutheran (Lu) blood group glycoprotein and the B-cell adhesion molecule (B-CAM) epithelial cancer antigen, have been recently cloned from human placenta and colon cancer HT29 cell line, respectively. Although amino acid sequences deduced from cDNA analysis suggested that B-CAM should represent an abridged form of the Lu glycoprotein lacking the last 40...
متن کاملThe Lutheran blood group glycoprotein, another member of the immunoglobulin superfamily, is widely expressed in human tissues and is developmentally regulated in human liver.
Glycoproteins expressing the Lutheran blood group antigens were isolated from human erythrocyte membranes and from human fetal liver. Amino acid sequence analyses allowed the design of redundant oligonucleotides that were used to generate a 459-bp, sequence-specific probe by PCR. A cDNA clone of 2400 bp was isolated from a human placental lambda gt 11 library and sequenced, and the deduced amin...
